Sheathless elasto-inertial focusing of sub-25 nm particles in straight microchannels

Nanoscale biological particles, such as lipoproteins (10-80 nm) or extracellular vesicles (30-200 nm), play pivotal roles in health and disease, including conditions like cardiovascular disorders and cancer. Their effective analysis is crucial for applications in diagnostics, quality control, and nanomedicine development. While elasto-inertial focusing offers a powerful method to manipulate particles without external fields, achieving consistent focusing of nanoparticles (<500 nm) has remained a challenge. In this study, we experimentally demonstrate elasto-inertial focusing of nanoparticles as small as 25 nm using straight high-aspect-ratio microchannels in a sheathless flow. Systematic investigations reveal the influence of channel width, particle size, viscoelastic concentration, and flow rate on focusing behavior. Additionally, through numerical simulations and experimental validation, we provide insights into particle migration dynamics and viscoelastic forces governing nanoparticle focusing. Finally, we successfully focus biological particles, including liposomes (90-140 nm), extracellular vesicles (100 nm), and lipoproteins (10-25 nm), under optimized conditions, showcasing potential applications in medical diagnostics and targeted drug delivery. These findings mark a significant advancement toward size-based high-resolution particle separation, with implications for biomedicine and environmental sciences.

Differential antibody dynamics to SARS-CoV-2 infection and vaccination

Optimal immune responses furnish long-lasting (durable) antibodies protective across dynamically mutating viral variants (broad). To assess robustness of mRNA vaccine-induced immunity, we compared antibody durability and breadth after SARS-CoV-2 infection and vaccination. While vaccination delivered robust initial virus-specific antibodies with some cross-variant coverage, pre-variant SARS-CoV-2 infection-induced antibodies, while modest in magnitude, showed highly stable long-term antibody dynamics. Vaccination after infection induced maximal antibody magnitudes with enhanced longitudinal stability while infection-naive vaccinee antibodies fell with time to post-infection-alone levels. The composition of antibody neutralizing activity to variant relative to original virus also differed between groups, with infection-induced antibodies demonstrating greater relative breadth. Differential antibody durability trajectories favored COVID-19-recovered subjects with dual memory B cell features of greater early antibody somatic mutation and cross-coronavirus reactivity. By illuminating an infection-mediated antibody breadth advantage and an anti-SARS-CoV-2 antibody durability-enhancing function conferred by recalled immunity, these findings may serve as guides for ongoing vaccine strategy improvement.

Memory B cell repertoire for recognition of evolving SARS-CoV-2 spike

Memory B cell reserves can generate protective antibodies against repeated SARS-CoV-2 infections, but with an unknown reach from original infection to antigenically drifted variants. We charted memory B cell receptor-encoded monoclonal antibodies (mAbs) from 19 COVID-19 convalescent subjects against SARS-CoV-2 spike (S) and found 7 major mAb competition groups against epitopes recurrently targeted across individuals. Inclusion of published and newly determined structures of mAb-S complexes identified corresponding epitopic regions. Group assignment correlated with cross-CoV-reactivity breadth, neutralization potency, and convergent antibody signatures. mAbs that competed for binding the original S isolate bound differentially to S variants, suggesting the protective importance of otherwise-redundant recognition. The results furnish a global atlas of the S-specific memory B cell repertoire and illustrate properties conferring robustness against emerging SARS-CoV-2 variants.

Integrated Cognitive Assessment: Speed and Accuracy of Visual Processing as a Reliable Proxy to Cognitive Performance

Various mental disorders are accompanied by some degree of cognitive impairment. Particularly in neurodegenerative disorders, cognitive impairment is the phenotypical hallmark of the disease. Effective, accurate and timely cognitive assessment is key to early diagnosis of this family of mental disorders. Current standard-of-care techniques for cognitive assessment are primarily paper-based, and need to be administered by a healthcare professional; they are additionally language and education-dependent and typically suffer from a learning bias. These tests are thus not ideal for large-scale pro-active cognitive screening and disease progression monitoring. We developed the Integrated Cognitive Assessment (ICA), a 5-minute computerized cognitive assessment tool based on a rapid visual categorization task, in which a series of carefully selected natural images of varied difficulty are presented to participants. Overall 448 participants, across a wide age-range with different levels of education took the ICA test. We compared participants ICA test results with a variety of standard pen-and-paper tests that are routinely used to assess cognitive performance. ICA had excellent test-retest reliability, and was significantly correlated with all the reference cognitive tests used here, demonstrating ICAs ability as one unified test that can assess various cognitive domains.
