RhythmWriter: GPU-accelerated Contractility Quantification for Large Datasets

Abstract

Human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) have been considered optimal model systems for use in cardiac safety testing, yet their adoption remains low, largely because of technical limitations in the analysis methodology. RhythmWriter (RW) is an open-source GPU plugin for ImageJ developed to quantify contractile motion over extended periods of time. RW compensates for baseline drift, is time-scale invariant and is capable of very rapid data analysis. As a consequence, RW is capable of generating simultaneous high-resolution contraction restitution curves from multiple aggregates of hiPSC-CMs derived from premature-stimulus (S1S2) and dynamic-ramp restitution protocols. The aggregates exhibit brief refractory periods, which allow the induction of S2 beats at coupling intervals down to 130 ms and 1:1 entrainment during periodic stimulation down to a cycle length of 245 ms. Simulations confirm the ability of RW to track programmed contractile strength and stimulation intervals even at rapid pacing rates. This makes possible, for the first time, high-throughput functional assessment of hiPSC-CM aggregates for contractility drug screening and maturation.

Sustained Maternal Cortisol Elevation Links Metabolic Stress to Fetal Growth Restriction in a Guinea Pig Model of Undernutrition

Fetal growth restriction (FGR) resulting from maternal undernutrition is commonly attributed to insufficient nutrient substrate delivery to the fetus. However, the contribution of maternal physiological stress responses to this outcome remains poorly characterized. Using the well-established guinea pig maternal nutrient restriction (MNR) model, we conducted a comprehensive retrospective analysis to interrogate the hypothesis that maternal stress physiology actively mediates fetal growth outcomes beyond simple substrate limitation. Maternal salivary and serum cortisol were significantly elevated in MNR dams throughout pregnancy despite no baseline differences at conception, indicating that hypothalamic-pituitary-adrenal (HPA) axis activation was specifically triggered by the combined physiological demands of pregnancy and nutritional constraint. The sustained cortisol elevation was accompanied by acute metabolic perturbations at mid-pregnancy, including hypoglycemia and depleted circulating lipids. While most acute metabolic disturbances resolved by near-term, cortisol remained elevated, and MNR fetuses exhibited parallel cortisol elevation alongside sustained growth restriction. Maternal weight divergence emerged only in late gestation and was driven primarily by reduced conceptus weight. Maternal organ analysis revealed selective hepatic enlargement with apparent lipid accumulation in MNR dams near-term. Overall, these findings help to reframe nutrition-related FGR as a condition in which maternal adaptive stress responses, reduced nutritional components and placental insufficiency are mechanistically intertwined, with important implications for the design of effective interventions beyond simply nutrient supplementation.

Dietary phosphorus restriction enables multigenerational breeding of α-Klotho-deficient rats

Severe -Klotho deficiency causes early mortality and reproductive impairment, limiting the maintenance and physiological analysis of rat models. We generated rats carrying a CRISPR/Cas9-mediated -Klotho deletion spanning exons 1-5 and evaluated dietary phosphorus restriction primarily in the Wistar-Imamichi background. Maternal low-phosphorus (LP) feeding from mating through lactation was associated with higher overall survival to weaning (92.6% versus 85.8% under normal-phosphorus conditions) and a higher proportion of KO animals among weaned survivors (34/113 versus 23/145). Maternal LP feeding also reduced plasma phosphorus in KO pups. Continued LP feeding after weaning supported growth, gonadal development, and natural KO x KO breeding. Across 15 litters, 74 of 85 offspring survived to weaning, and KO colonies were maintained to at least the fifth generation. We additionally characterized 24-h metabolic rhythms in LP-maintained KO animals using indirect calorimetry. These profiles remained heterogeneous under a light-dark cycle. In conclusion, dietary phosphorus restriction provides a reliable strategy for the multigenerational maintenance of severe -Klotho-deficient rats, establishing a maintainable model to investigate residual metabolic and physiological phenotypes.

Reduced physiological reserve and a blunted response to acute exercise in adults with cerebral palsy

Cerebral palsy (CP) is an early-onset neurodevelopmental condition associated with metabolic impairment and an increased risk of non-communicable diseases across the lifespan. However, the basis of these changes remains largely unknown. This study provides a comprehensive cardiometabolic and physical function profile of 23 adults with CP relative to 33 typically developing (TD) adolescents and 61 TD adults. Compared to TD individuals, adults with CP exhibited reduced cardiorespiratory fitness, smaller cardiac dimensions, and lower muscle mass. Circulating metabolomic and proteomic profiling identified a distinct molecular signature in CP, including altered tricarboxylic acid (TCA) cycle metabolites, and elevated myostatin, matrix-related proteins and pro-inflammatory cytokines. We further tested the capacity for physiological and molecular adaptation using an acute bout of endurance exercise. In response to exercise, individuals with CP exhibited an attenuated response compared to TD groups, characterized by a blunted lactate response and lower venous oxygen levels. TD participants also increased canonical exercise-responsive factors, including NADK, IL-8, ANGPTL1 and GDF15, whereas these responses were attenuated in CP. These novel findings define a distinct cardiometabolic profile in adults with CP at rest and reveal an attenuated molecular response to exercise. These data carry implications for targeted interventions and prevention strategies across the lifespan.
