Extracellular Vesicle-delivered Bone Morphogenetic Proteins: A novel paracrine mechanism during embryonic development

Abstract

Morphogens including Wnt, Hedgehog and BMP proteins are essential during embryonic development and early induction of organ progenitors. Besides free diffusion to form signalling gradients, extracellular vesicle- (EV-) mediated morphogen transport was identified as a central mechanism for Wnt- and Hh-signalling. Here, we investigated EVs isolated from whole zebrafish embryos as a potential morphogen transport mechanism. Inhibition of EV-secretion during development leads to severe dorsalization phenotypes, reminiscent of disrupted BMP-signalling. Subsequently, we found that EVs isolated from zebrafish embryos at bud stage contain biologically active BMP2/4 protein. Embryos with inhibited EV secretion display reduced Smad1/5/9-phosphorylation and downstream gene expression activity. We further show that BMP-containing EVs are secreted by endodermal cells in vitro, and inhibition of endodermal-EV release in vivo causes signs of BMP signalling loss. Our data provides evidence that establishes the transport of BMP2/4 by EVs as an essential but so far undiscovered mechanism in developmental morphogenesis.

Optimizing the efficiency of assisted reproduction procedures in the common marmoset (Callithrix jacchus) improved IVF outcomes

The marmoset has emerged as a model species for genetic studies of complex neurodevelopmental conditions because of their brain development, behavior, communication, and compact size. IVF has been used to generate wild-type and transgenic marmoset embryos, but inefficiency of IVF in the marmoset compared to other primates poses challenges for genetic manipulation. To address this limitation, we analyzed the association between variables of age, number of cycles, and progesterone levels for IVF and embryology outcomes. Longitudinal data collected over 2 years indicated that the progesterone levels 24 hours after Cloprostenol sodium (Estrumate) administration, the number of days of ovarian stimulation during an IVF cycle, and the number of oocyte retrievals that an animal had undergone all influenced IVF cycle outcomes. Higher serum progesterone levels at cycle start were negatively correlated with the likelihood of fertilized eggs progressing to the cleavage stage of embryo development. Additionally, a shorter duration of ovarian stimulation (ranging 7 to 14 days of medication) coupled with a GnRH antagonist correlated with an increased percentage of fertilized eggs. We observed a trend that a shorter hormonal stimulation cycle length was associated with a higher percentage of embryos reaching the cleavage stage. The optimization of stimulation cycle protocol and embryology has led to a high percentage of implantation after embryo transfer regardless of whether the embryo was fresh or cryopreserved. These findings suggest that shorter IVF cycles and a progesterone level <10 ng/mL at cycle start were correlated with an increased likelihood of a cleavage stage embryo development.

Yap is a key regulator of tracheoesophageal separation during foregut development

Separation of the anterior foregut into the esophagus and trachea involves dynamic cellular movement that requires the coordinated activity of multiple signaling pathways and transcription factors. Disruption of this process can result in esophageal atresia (EA) with or without tracheoesophageal fistula (TEF), manifesting in five major anatomical types (Types A-E). Here, we examine the role of the Hippo pathway effector Yes-associated protein (Yap) in foregut separation. Conditional deletion of Yap in the dorsal and medial portions of the anterior foregut endoderm leads to the formation of Type A EA/TEF, isolated EA. This separation defect only occurs in Sox2-CreER;Yaploxp/loxp but not in Shh-Cre;Yaploxp/loxp mutants. Our findings further reveal that Yap mutants exhibit a reduction in Sox2+ esophageal progenitor cells accompanied by the absence of the middle esophageal segment. Combined deletion of Yap and Taz (transcriptional coactivator with PDZ-binding motif) also results in isolated EA accompanied by severe craniofacial abnormalities. Therefore, loss of Yap causes a distinct Type A EA/TEF, which differs from existing mouse models of EA/TEF that predominantly exhibit the Type C phenotype. Our findings also suggest that epithelial cells at the dorsal-ventral boundary play a critical role in tracheoesophageal separation.

Tissue geometry patterns epidermal stem cell fate in murine interfollicular skin

How tissues maintain spatial patterns of stem cell renewal and differentiation despite continuous cellular turnover remains poorly understood. Here, we show that microscopic undulations of the dermal-epidermal junction (DEJ) are associated with distinct stem cell behaviors in the mouse epidermis. Using longitudinal intravital imaging and quantitative analysis of cell morphology and lineage dynamics, we find that local DEJ curvature predicts the subsequent fate of daughter cells up to three days before mother cell division. Cells residing in concave valleys preferentially produce proliferating descendants, whereas convex ridges are associated with increased differentiation. These spatial biases are reflected in the expression of stem- and differentiation-associated markers and accumulate into differences in long-term clonal expansion. Remarkably, DEJ geometry remains stable for weeks, persisting across multiple generations of epidermal cells. Moreover, human epidermal equivalents grown on devitalized dermis recapitulate the preferential retention of basal layer cells in valleys, indicating that living dermal cells are not required for this spatial organization. Our findings suggest that stable tissue architecture can pattern stem cell fate probabilities across generations, contributing to functional stem cell heterogeneity.
