An interferon-like off-switch in invertebrates: Vago4 downregulates Domeless to restrain JAK/STAT signaling

Whether invertebrates possess a functional interferon (IFN)-like system remains controversial, as canonical IFN ligands and their receptors are absent from invertebrate genomes. Although Vago proteins in Drosophila and shrimp are compelling functional analogs of IFN, direct evidence for a canonical cytokine-receptor axis and, importantly, for how such signaling is restrained to prevent excessive immune activation, has been lacking. Here, we identify Vago4 as a bona fide ligand of Domeless, the canonical type I cytokine receptor upstream of JAK/STAT signaling. Vago4 binds the extracellular CBM1 and CBM2 domains of Domeless, triggering receptor endocytosis followed by lysosomal trafficking and K27-linked ubiquitination-dependent proteasomal degradation. This ligand-induced receptor downregulation serves as a negative feedback mechanism that restrains excessive JAK/STAT activation. During acute Vibrio infection, Vago4 silencing accelerated shrimp mortality, whereas recombinant Vago4 alleviated inflammatory lesions and oxidative stress, enhanced phagocytosis, and improved survival, phenocopying JAK or STAT knockdown. Thus, by downregulating Domeless, Vago4 limits immunopathology and defines an IRF-Vago4-Domeless-JAK/STAT IFN-like axis in invertebrates, offering new insight into the evolution of IFN-mediated immune regulation.

The Red Rice Bran Extract (RRBE) Mitigates Photoaging by Targeting Mitochondrial Oxidative Stress and Regulating Thermal Damage Responses

The photoprotective efficacy of natural skin-active complexes is well recognized, yet their ap-plication is often hindered by the challenge of deciphering their complex, multi-component, and multi-target mechanisms. To bridge the gap between established phenotypes and molecular mechanisms, we developed an AI-driven integrated platform that combines phytochemical profiling, network pharmacology, and deep learning-based target prediction with rigorous biophysical validation. We applied this platform to investigate RRBE, a bioactive complex refined from red rice bran extract. In vivo clinical studies confirmed that RRBE significantly accelerates the resolution of UV-induced erythema, while cellular and 3D tissue models demonstrated robust suppression of oxidative stress and DNA damage responses. To decode its material basis, the platform deconstructed RRBE into 10 distinct chemical modules. Leveraging our SCOPE-DTI deep learning model for global target prediction, we identified flavonoids (Module 1) and phenolic acids (Module 10) as the primary bioactive drivers relevant to photoprotection. These computational predictions were structurally supported by molecular docking and definitively validated in a physiological environment via Cellular Thermal Shift Assay-Mass Spectrometry (CETSA-MS). Mechanistically, RRBE functions through a synergistic polypharmacology: (1) Module 1 components, represented by procyanidin B2, target NDUFA7 to stabilize mitochondrial function and mitigate ROS; (2) Module 10 components, exemplified by caffeic acid, ferulic acid, and p-coumaric acid, engage FKBP11 and HSP90AA1 to regulate protein homeostasis and stress responses. This work not only deciphers the polypharmacological basis of RRBEs photoprotective action but also validates a scalable "AI-guided, cell-validated" discovery pipeline, offering a rational paradigm for uncovering the protective benefits of complex natural extracts.

Enhanced Functional Potential of Pseudogene-associated lncRNA Genes in Mammals

The functional significance of long non-coding RNAs (lncRNAs) remains a subject of debate, largely due to the complexity and cost associated with their validation experiments. However, emerging evidence suggests that pseudogenes, once viewed as genomic relics, may contribute to the origin of functional lncRNA genes. In this study spanning eight species, we systematically identified pseudogene-associated lncRNA genes using our PacBio long-read sequencing data and published RNA-seq data. Our investigation revealed that pseudogene-associated lncRNA genes exhibit heightened functional attributes compared to their non-pseudogene-associated counterparts. Notably, these pseudogene-associated lncRNAs show protein-binding proficiency, positioning them as potent regulators of gene expression. In particular, pseudogene-associated sense lncRNAs retain protein-binding capabilities inherited from parent genes of pseudogenes, thereby demonstrating greater protein-binding proficiency. Through detailed functional characterization, we elucidated the unique advantages and conserved roles of pseudogene-associated lncRNA genes, particularly in the context of gene expression regulation and DNA repair. Leveraging cross-species expression profiling, we demonstrated the prominent contribution of pseudogene-associated lncRNA genes to aging-related transcriptome changes across nine human tissues and eight mouse tissues. Overall, our findings demonstrate enhanced functional attributes of pseudogene-associated lncRNA genes and shed light on their conserved and close association with aging.

Single-Cell Map of Dynamic Multicellular Ecosystem of Radiation-Induced Intestinal Injury

Intestine is a highly radiation-sensitive organ that could be injured during the radiotherapy for abdominal or pelvic cavity tumors. However, the dynamic change of the intestinal microenvironment related to radiation-induced intestine injury (RIII) is still unclear. Using single-cell RNA sequencing, we pictured a dynamic landscape of the intestinal microenvironment during RIII and regeneration. We showed that the multicellular ecosystem of intestine exhibited heterogeneous radiosensitivities. We revealed the distinct dynamic patterns of three subtypes of intestinal stem cells (ISCs), and the cellular trajectory analysis suggested a complex interconversion pattern among them. For the immune cells, we found that Ly6c+ monocytes can give rise to both pro-inflammatory macrophages and resident macrophages after RIII. Besides, through cellular communication analysis, we identified a positive feedback loop between the macrophages and endothelial cells, which could amplify the inflammatory response induced by radiation. Overall, our study provides a valuable single-cell map of the dynamic multicellular ecosystem during RIII and regeneration, which may facilitate the understanding of the mechanism of RIII.

Impulsivity and Thought Suppression in Behavioral Addiction: Associated Neural Connectivity and Neural Networks

Impulsivity and thought suppression are two psychological traits that have great variation in healthy population. In extreme cases, both are closely related to mental illness and play an important role in behavioral addiction. We have known the role of the top-down mechanism in impulsivity and thought suppression, but we do not know how the related neural nuclei are functionally connected and interact with each other. In the study, we selected excessive internet users (EIU) as our target population and investigated the relationship between thought suppression and impulsivity in the following aspects: their correlations to psychological symptoms; the associated neural networks; and the associated brain morphometric changes. We acquired data from 131 excessive internet users, with their psychological, resting-state fMRI and T1-MRI data collected. With the whole brain analysis, graph theory analysis, replication with additional brain atlas, replication with additional MRI data, and analysis of brain structure, we found that: (i) implusivity and thought suppression shared common neural connections in the top-down mechanism; (ii) thought suppression was associated with the neural network that connected to the occipital lobe in the resting-state brain but not the morphometric change of the occipital lobe. The study confirmed the overlap between impulsivity and thought suppression in terms of neural connectivity and suggested the role of thought suppression and the occipital network in behavioral addiction. Studying thought suppression provided a new insight into behavioral addiction research. The neural network study helped further understanding of behavioral addiction in terms of information interaction in the brain.
