Integrative Analysis the characterization of peroxiredoxins in pan-cancer

Peroxiredoxins (PRDXs) are antioxidant enzymes protein family members that involves the process of several biological functions, such as differentiation, cell growth. Considerable evidence demonstrates that PRDXs play critical roles in the occurrence and development of carcinomas. However, a systematic analysis of PRDXs in cancers is deficiency. Therefore, we perform a comprehensive analysis of PRDXs in 33 cancer types including mRNA expression profiles, genetic alterations, methylation, prognostic values, potential biological pathways and target drugs. Moreover, we validated that PRDX6 could regulate cancer cell proliferation via JAK2-STAT3 pathway and involve into the process of cell cycle in bladder cancer.

Regular Tai Chi Practice Is Associated with Improved Memory as well as Structural and Functional Integrity of the Hippocampal Formation in the Elderly

ObjectiveThe current study aimed at determining effects of Tai Chi as an example of a combined motor-cognitive exercise relative to regular walking as an example of an exercise without cognitive demands on cognitive functioning and the functional and structural integrity of the brain in the elderly. MethodsHealthy elderly women with at least 6 years of regular Tai Chi or brisk walking exercise were recruited and underwent cognitive assessment via the Montreal Cognitive Assessment and brain structural and resting state functional MRI assessments. ResultsEpisodic memory in Tai Chi group was superior to that of the walking group; (2) higher gray matter density in inferior and medial temporal regions, including the hippocampal formation; (3) higher ReHo in temporal regions, specifically the fusiform gyrus and hippocampal formation (4) significant partial correlations were found between the gray matter density of the left hippocampus and episodic memory in the whole sample (5) significant partial correlations were observed between the ReHo in left hippocampus, left parahippocampal, left fusiform and delayed memory task was observed among all subjects. ConclusionThe present study suggest that long-term Tai Chi practice may improve memory performance via remodeling structure and the function of the hippocampal formation.

A Targeted Vaccine against COVID-19: S1-Fc Vaccine Targeting the Antigen-Presenting Cell Compartment Elicits Protection against SARS-CoV-2 Infection

Vaccination efficacy is enhanced by targeting the antigen-presenting cell compartment. Here, we show that S1-Fc antigen delivery targeting the Fc{gamma}R+ antigen-presenting cell compartment elicits anti-SARS-CoV-2 S1-antigen specific IgG production in vivo exerting biologically functional and protective activity against live virus infection, assessed in a stringent experimental virus challenge assay in vitro. The S1-domain of the SARS-CoV-2 spike protein was genetically fused to a human immunoglobulin Fc moiety, which contributes to mediate S1-Fc cellular internalization by Fc{gamma}R+ antigen-presenting cells. Immediately upon administration intramuscularly, our novel vaccine candidate recombinant rS1-Fc homes to lymph nodes in vivo where Fc{gamma}R+ antigen-presenting cells reside. Seroconversion is achieved as early as day 7, mounting considerably increased levels of anti-S1 IgGs in vivo. Interestingly, immunization at elevated doses with non-expiring S1-Fc encoding dsDNA favors the education of a desired antigen-specific adaptive T cell response. However, low-dose immunization, safeguarding patient safety, using recombinant rS1-Fc, elicits a considerably elevated protection amplitude against live SARS-CoV-2 infection. Our promising findings on rS1-Fc protein immunization prompted us to further develop an affordable and safe product for delivery to our communities in need for COVID-19 vaccinations.
