Vacuolar type H+ ATPase is involved in stress responses in Leishmania mexicana by regulating the lysosomal pH

Vacuolar H+ ATPases (v-ATPases) are conserved proton pumps that support diverse biological functions through acidification of cellular organelles. The protozoan parasite Leishmania requires its v-ATPase for survival in the sand fly vector and mammalian host, but genetic mutants remain viable in vitro. To gain further insight into this conditionally lethal phenotype, we first mapped organellar localization of the v-ATPase by co-localisation imaging of fluorescently tagged v-ATPase subunits and organelle markers. The v-ATPase signal was strongest in the flagellar pocket region, consistent with enrichment in the contractile vacuole complex (CVC). To define the conditions that require a functional v-ATPase, deletion mutants were exposed to different stresses (pH, temperature, osmolality, dense culture). All tested deviations from standard culture conditions affected the mutants' growth rate, viability or both. Despite differences in phenotype severity, all stressors triggered the formation of a large autolysosome, positive for the autophagy marker protein ATG8 and the lysosomal enzyme cysteine peptidase A, indicating an arrest at the final step of autophagy. Measurements with the pH sensor pHLuorin2 showed that the luminal pH of the lysosomes was 5.6 in unperturbed promastigotes and 7.1 in v-ATPase mutants. These data support a canonical function for the Leishmania v-ATPase in lysosome acidification and autophagy, which is essential for parasite differentiation, and identify the poorly characterized Leishmania CVC as another major site of v-ATPase concentration.

MARINER: a surround visual stimulator for vision research in aquatic animals

Aquatic vertebrates are increasingly used in neuroscience research, yet underwater visual stimulation remains a challenge. Commonly used monochromatic stimuli have been shown to be inadequate to activate many visual neurons properly, and underwater refraction artifacts are prone to ruin stimulus designs. Here, we present MARINER - a visual stimulator, which remedies these issues and integrates concurrent behavioral and neurophysiological two-photon calcium imaging recordings. MARINERs full-field visual stimulation combined with receptive field mapping reveals that the visual field of larval zebrafish is larger than previously thought, extending almost down below the fish, and is spatially biased to better utilize motion content in naturalistic visual scenes. Using chromatic motion nulling, we further show that behavioral responses and task-associated sensory neurons are colorblind for "red" and "green" during the larvas optokinetic response. The MARINER stimulator facilitates naturalistic stimulation and faithful presentation of colored visual underwater stimuli for small aquatic species.

OSBP-mediated PI(4)P-cholesterol exchange at endoplasmic reticulum-secretory granule contact sites controls insulin secretion

Insulin secretion is the process whereby insulin-containing granules fuse with the plasma membrane of pancreatic {beta}-cells. Exocytosis is preceded by cargo loading and granule biogenesis at the Golgi, followed by maturation and transport of the secretory granules; processes that require modification of both the protein and lipid composition of the granules. Here, we show that insulin-containing secretory granules form physical contacts with the endoplasmic reticulum. The lipid exchange protein OSBP dynamically redistributes to ER-SG contacts in a process regulated by Ca2+ and cytosolic pH, and contributes to cholesterol loading of the granules. This process depends on granular PI(4)P and ER-localized VAPs, and is positively regulated by granular PI4-kinases and negatively regulated by granule-localized Sac2. Loss of Sac2 results in excess accumulation of cholesterol on insulin granules that is normalized when OSBP expression is reduced, and both acute inhibition and siRNA-mediated knockdown of OSBP suppresses glucose-stimulated insulin secretion without affecting insulin production or intracellular Ca2+ signaling. In conclusion, we show that lipid exchange at ER-granule contact sites is involved in the exocytic process, and propose that these contacts act as reaction centers with multimodal functions during insulin granule maturation.

Number of days required to estimate objectively measured physical activity constructs in different age groups

PurposeThe present study aims to estimate the minimum number of accelerometer measurement days needed to estimate habitual physical activity (PA) among 6, 18 and 30 years old participants, belonging to three population-based Brazilian birth cohorts. Method: PA was assessed by triaxial wrist worn GENEActiv accelerometers for 4-7 consecutive days, including at least one weekend day. Accelerometer raw data were analyzed with R-package GGIR. Description of PA measures (overall PA, MVPA, LPA) between weekdays and weekend days were conducted, and statistical differences were tested with chi-squared and Kruskal-Wallis tests. Intraclass Reliability Coefficient (IRC) was applied through the Spearman Brown Formulae to test reliability of different number of days of accelerometer use. Results: Differences between week and weekend days regarding LPA, MVPA and Overall PA, were only observed among 30-year-olds. More MVPA (p=0.006) and Overall PA (p<0.001) were performed on week days. The IRC coefficients ranged from 0.44 to 0.83 in children and from 0.54 to 0.88 in adults. Conclusion: In conclusion, our results show that between four and six measurement days are needed to achieve good reliability in the analyzed participants, depending on the PA construct evaluated (MVPA, LPA or overall PA).
