Integration of auxin and cytokinin signalling in Arabidopsis root meristem maintenance through the secreted peptide CLE40

Plant meristems perpetually generate new cells for the initiation, growth and differentiation of various cell types and organs. CLAVATA3/EMBRYO SURROUNDING REGION-RELATED (CLE) genes encode plant specific peptides exerting indispensable functions in a multitude of these developmental processes. Previously, we have described a CLE40 signaling module that regulates stemness in the distal root meristem (DRM). Here, we show that CLE40 is additionally needed to maintain the proximal root meristem (PRM). We demonstrate that in cle40 mutants, meristematic cells differentiate prematurely, resulting in shortened PRMs and ultimately, reduced root growth. Furthermore, we link these effects to increased cytokinin signaling that can be circumvented by preventing the perception of cytokinin. We highlight how increased cytokinin signaling leads to a change in auxin distribution mediated by SHORT HYPOCOTYL 2 (SHY2), which is ectopically activated if CLE40 is not active. Finally, we propose that the receptor BARELY ANY MERISTEM 1 (BAM1), but not its homolog BAM2, perceives CLE40 in the PRM.

Lossless Single-Molecule Counting To Absolute QuantifyProteoforms

A novel immunoassay, termed Protein Interaction Coupling (PICO), is introduced to deliver unequivocal, reference-free quantification of proteoforms - absolute quantification. PICO employs a compartmentalized, homogeneous single-molecule assay with a lossless and highly sensitive signal generation, capable of detecting down to a few molecules per reaction. Additionally, it utilises a background-free, digital enumeration principle, known as the decouplexing. PICO is presented as exact mathematical theories, providing a theoretical comprehension of its chemistry. Consequently, PICO demonstrates absolute quantification, as exemplified with recombinant and non-recombinant ErbB2 and multi-tagged peptide rTRX targets, validating absolute quantification against internal and external references in both analytical and cellular matrices. Furthermore, PICO enables combinatorial multiplexing (cplex), a readout between any two antibodies, demonstrated by an 8-plex antibody, 12-cplex PICO, measuring functional changes of ErbB pathway upon mock and dactolisib treatment delivering absolute quantitative cellular stoichiometry. PICO possesses immense potential for versatile, standardized, and accurate protein measurements, offering insights into physiological and perturbed cellular processes.

The cell fate controlling CLE40 peptide requires CNGC9 to trigger highly localized Ca2+ transients in Arabidopsis thaliana root meristems

Communication between plant cells and their biotic environment is largely dependent on the function of plasma membrane localized receptor-like kinases (RLKs). Major players in this communication within root meristems are secreted peptides, including CLAVATA3/EMBRYO SURROUNDING REGION40 (CLE40). In the distal root meristem, CLE40 acts through the receptor like kinase (RLK) ARABIDOPSIS CRINKLY4 (ACR4) and the leucine-rich repeat (LRR) RLK CLAVATA1 (CLV1) to promote cell differentiation. In the proximal meristem, CLE40 signalling requires the LRR receptor-like protein (RLP) CLAVATA2 (CLV2) and the membrane localized pseudokinase CORYNE (CRN), and serves to inhibit cell differentiation. The molecular components that act immediately downstream of the CLE40-activated receptors are not yet known. Here we show that active CLE40 signalling triggers the release of intracellular Ca2+ leading to increased cytosolic Ca2+ concentration ([Ca2+]cyt) in a subset of proximal root meristem cells. This rise in [Ca2+]cyt depends on the CYCLIC NUCLEOTIDE GATED CHANNEL 9 (CNGC9), CLV1, the CLV1-related BARELY ANY MERISTEM1 (BAM1), CLV2 and CRN. The precise function of changes in [Ca2+]cyt are not yet known, but might form a central part of a fine-tuned response to CLE40 peptide that serves to integrate root meristem growth with stem cell fate decisions and initiation of lateral root primordia.

The IDA cell separation pathway connects developmental and defense responses

The abscission of floral organs and emergence of lateral roots in Arabidopsis is regulated by the peptide ligand INFLORESCENCE DEFICIENT IN ABSCISSION (IDA) and the receptor protein kinases HAESA (HAE) and HAESA-LIKE 2 (HSL2). During these cell separation processes, the plant induces defense-associated genes to protect against pathogen invasion. However, the molecular coordination between abscission and immunity has not been thoroughly explored. Here we show that IDA induces a release of cytosolic calcium ions (Ca2+) and apoplastic production of reactive oxygen species, which are signatures of early defense responses. In addition, we find that IDA promotes late defense responses by the transcriptional upregulation of genes known to be involved in immunity. When comparing the IDA induced early immune responses to known immune responses, such as those elicited by flagellin22 treatment, we observe both similarities and differences. We propose a molecular mechanism by which IDA promotes signatures of an immune response in cells destined for separation to guard them from pathogen attack.
