VTA GABA Cell Bipotential Induction of iLTP or iLTD Synaptic Plasticity is Input Selective, where iLTD is Uniquely Eliminated by Cocaine.

The ventral tegmental area (VTA) is a key reward circuit hub, implicated in drug seeking and addictive behaviors. The VTA contains dopaminergic and GABAergic neurons that both play roles in reward prediction, aversion, motivated reward behavior, etc. Synaptic plasticity, including VTA excitatory and inhibitory long-term potentiation (LTP/iLTP) and long-term depression (LTD/iLTD), are fundamentally involved in processing reward learning and memory, which is maladaptively altered by abused drugs mediating dependence induction. This report extends our prior research of the understudied VTA GABA cells and the rationale for their bipotential plasticity (iLTP or iLTD) capacity by optogenetic circuit level examination of their unique GABAergic inputs. In addition, we examine potential cocaine impact on both plasticity forms. Optogenetic activation of either lateral hypothalamus or rostromedial tegmental nucleus induced iLTP in VTA GABA cells, while optogenetic activation of local VTA GABAergic inputs induced iLTD. This suggests expression of bipotential plasticity is input specific, and highlights implications for each type of plasticity on reward signaling. Drug of abuse cocaine eliminated iLTD while sparing iLTP, suggesting selective impairment of local GABA signaling to VTA GABA cells by cocaine versus projection GABA signaling. This emphasizes potential differential VTA GABA cell plasticity in reward processing and the need to further examine cocaine impact on inhibitory signaling in addition to known impact on the dopaminergic system. By elucidating the circuit-dependence of plasticity type in VTA GABA cells and drug-induced impact, our research could potentially identify additional targets for therapeutic intervention of drug dependence via the GABAergic system.

Genetic and anatomical determinants of olfaction in dogs and wild canids

Understanding the anatomical and genetic basis of complex phenotypic traits has long been a challenge for biological research. Domestic dogs offer a compelling model as they demonstrate more phenotypic variation than any other vertebrate species. Dogs have been intensely selected for specific traits and abilities, directly or indirectly, over the past 15,000 years since their initial domestication from the gray wolf. Because olfaction plays a central role in critical tasks, such as the detection of drugs, diseases, and explosives, as well as human rescue, we compared relative olfactory capacity across dog breeds and assessed changes to the canine olfactory system resulting from domestication. We conducted a cross-disciplinary survey of olfactory anatomy, olfactory receptor (OR) gene variation, and OR gene expression in domestic dogs. Through comparisons to their closest wild canid relatives, the gray wolf and coyote, we show that domestic dogs might have lost functional OR genes commensurate with a documented reduction in nasal morphology during domestication. Critically, within domestic dogs alone, we found no genetic or morphological profile shared among functional or genealogical breed groupings, such as scent hounds, that might indicate evidence of any human-directed selection for enhanced olfaction. Instead, our results suggest that superior scent detection dogs likely owe their success to advantageous behavioral traits and training rather than an "olfactory edge" provided by morphology or genes.

Bacopa monnieri phytochemicals regulate fibroblast cell migration via modulation of focal adhesions

The Bacopa monnieri plant contains a large repertoire of active phytochemicals that have been used extensively in traditional medicine for the treatment of various complex diseases. More recently it has been shown to increase the wound healing rate in rats, though its mechanism of action is largely unknown. Here we investigated the cellular pathways activated by a methanol extract of Bacopa monnieri in human dermal fibroblasts, which play many critical roles in the wound healing program. Gene expression analysis revealed that Bacopa monnieri extract can enhance tissue repair by modulating multiple processes involved in the wound healing program such as migration, proliferation, and angiogenesis. We discovered that Bacopa monnieri extract can increase migration of fibroblasts via modulating the size and number of focal adhesions. Bacopa monnieri-mediated changes in focal adhesions are dependent on 5{beta}1 integrin activation and subsequent phosphorylation of Focal Adhesion Kinase (FAK). Altogether our results suggest that Bacopa monnieri extract could enhance the wound healing rate via modulating fibroblast migration in the wound bed.

Towards genetic modification of plant-parasitic nematodes: delivery of macromolecules to adults and expression of exogenous mRNA in second stage juveniles.

Plant-parasitic nematodes are a continuing threat to food security, causing an estimated 100 billion USD in crop losses each year. The most problematic are the obligate sedentary endoparasites (primarily root knot nematodes and cyst nematodes). Progress in understanding their biology is held back by a lack of tools for functional genetics: forward genetics is largely restricted to studies of natural variation in populations, and reverse genetics is entirely reliant on RNA interference. There is an expectation that the development of functional genetic tools would accelerate the progress of research on plant-parasitic nematodes, and hence the development of novel control solutions. Here, we develop some of the foundational biology required to deliver a functional genetic tool kit in plant-parasitic nematodes. We characterise the gonads of male Heterodera schachtii and Meloidogyne hapla in the context of spermatogenesis. We test and optimise various methods for the delivery, expression, and/or detection of exogenous nucleic acids in plant-parasitic nematodes. We demonstrate that delivery of macromolecules to cyst and root knot nematode male germlines is difficult, but possible. Similarly, we demonstrate the delivery of oligonucleotides to root knot nematode gametes. Finally, we develop a transient expression system in plant-parasitic nematodes by demonstrating the delivery and expression of exogenous mRNA encoding various reporter genes throughout the body of H. schachtii juveniles using lipofectamine-based transfection. We anticipate these developments to be independently useful, will expedite the development of genetic modification tools for plant-parasitic nematodes, and ultimately catalyze research on a group of nematodes that threaten global food security.

The Life History of Human Foraging: Cross-Cultural and Individual Variation

Human adaptation depends upon the integration of slow life history, complex production skills, and extensive sociality. Refining and testing models of the evolution of human life history and cultural learning will benefit from increasingly accurate measurement of knowledge, skills, and rates of production with age. We pursue this goal by inferring individual hunters of hunting skill gain and loss from approximately 23,000 hunting records generated by more than 1,800 individuals at 40 locations. The model provides an improved picture of ages of peak productivity as well as variation within and among ages. The data reveal an average age of peak productivity between 30 and 35 years of age, though high skill is maintained throughout much of adulthood. In addition, there is substantial variation both among individuals and sites. Within study sites, variation among individuals depends more upon heterogeneity in rates of decline than in rates of increase. This analysis sharpens questions about the co-evolution of human life history and cultural adaptation. It also demonstrates new statistical algorithms and models that expand the potential inferences drawn from detailed quantitative data collected in the field.
