Genomic instability and biofilm determinants in Streptococcus mutans: insights from a sequence-defined arrayed transposon library

Streptococcus mutans is a primary architect of dental caries, utilizing complex genetic networks to build resilient, acid-producing biofilms. While pooled screens (Tn-seq) have identified important fitness factors, they often fail to capture extracellular or moderate-effect determinants due to community-level masking. Therefore, to study biofilm phenotypes, we constructed a comprehensive arrayed library of 9,216 mutants and used Cartesian Pooling-Coordinate Sequencing (CP-CSeq) to establish a sequence-defined resource covering 51% of non-essential genes. By screening the entire collection in isolation, we identified several novel biofilm determinants, including the putative metal transporter SMU_635 and the glycosylation-associated protein SMU_2160. However, systematic whole-genome sequencing (WGS) of our hits revealed an interesting level of genomic instability: 25% of biofilm-defective mutants had undergone spontaneous recombination at the gtfBC locus, while 7% had lost the TnSmu1 element, an excision rate 1,000-fold higher than previously reported. While targeted mutagenesis confirmed that TnSmu1 loss does not impact biofilm integrity, the gtfBC deletions directly accounted for the most severe phenotypes, highlighting a systemic risk of misattributing gene functions to primary transposon insertions. Our findings provide a powerful new genetic resource for the S. mutans community while establishing a critical new standard: an arrayed library is only as defined as its underlying genome, making systematic genomic verification an essential prerequisite for accurate functional genomics. ImportanceStreptococcus mutans is a major human pathogen responsible for dental caries, a global public health challenge driven in part by the organisms ability to form resilient, acidogenic biofilms. While traditional pooled genetic screens have identified many fitness factors, they often fail to capture extracellular or moderate-effect determinants because neighboring healthy bacteria can mask these defects. This work provides the scientific community with a sequence-defined arrayed mutant library, an essential resource for dissecting the individual contributions of genes to biofilm integrity in isolation. Beyond identifying well-known machinery, this study uncovers novel determinants, including the putative metal transporter SMU_635 and the putative glycosylation-associated protein SMU_2160. Crucially, the discovery of pervasive genomic instability within the library, specifically at the gtfBC and TnSmu1 loci, reveals a systemic risk in functional genomics: the potential to misattribute phenotypes to primary mutations when the underlying background has undergone large-scale rearrangements. By establishing systematic whole-genome verification as a necessary standard, this research ensures that the identification of future therapeutic targets is built upon a verified genetic foundation.

Assessment of Water Stress Tolerance in Mungbean Induced by Polyethyline Glycol

Water scarcity is a common hindrance of crop production in the globe especially in dry regions due to low precipitation along with high air temperature. Mungbean is a valuable pulse crop faced considerable amount of water shortage during its growing periods (March-May). Thus, the study was carried out to find out the variability and diversity present in the mungbean genotypes based on genetic variation of germination and seedling growth traits induced by polyethyline glycol (PEG-6000) stress (-0.7, -1, -2 and -4 bar). Seeds of each genotype was placed in a 9 cm diameter petridish containing sand bed that moistened by respective prepared PEG solutions and allowed to grow up to 10 days after placement. The result showed that the phenotypic coefficient of variation was significantly greater than the genotypic coefficient of variation for the entire studies traits, representing that these traits was influence by the environmental factors. The heritability showed moderate to high (24.26%-99.19%) under the observed traits indicating that these traits were under the control of additive genetic effects. The clustering pattern exhibited that cluster III were the largest ones with eleven genotypes. In case of contribution of different traits, the first five components were found to contribute for 98.19 % of the total variation while only first two components contributed for 82.12 % variation. The greater inter-cluster distance was found among the cluster I with II, III, IV and V. The maximum intra-cluster distance was observed in the cluster II (0.12). The genotypes in the cluster V recorded for greater performance by giving the highest mean value for germination, coefficient of germination, speed of germination, vigor index, shoot length and root length. This mean that genotype under cluster V have any gene or mechanism which responsible for tolerant higher level of water stress. The results of the study can be caring for establishing assortment criteria for water stress tolerant mungbean genotypes under breeding program.

Age analysis and growth patterns of Tenualosa ilisha using otolith examination and length-weight relationships from different regions of Bangladesh

The assessment of age and growth patterns provides crucial information on numerous aspects of the population dynamics in fish, which can, in turn, be used to determine a sustainable fishing rate and inform effective resource management practices. However, studies on the age and growth pattern of Tenualosa ilisha (commonly referred to as hilsa) are plagued by a lack of essential information; addressing this data gap is the aim of the present study. Six diverse habitats across Bangladesh were chosen as sampling sites for hilsa collection. For age determinations, the lunar rings in the otolith of the hilsa fish that are periodically-deposited in accordance with the lunar cycle were used to reflect 14-day increments of time. The length and weight of each fish were recorded prior to otolith extraction. The resulting otoliths were polished to enable the visualization and quantification of their lunar rings with a high-magnification microscope. Except for the Kali River and Gaglajur Haor samples, the age of the fish correlated strongly with both their length and weight (r > 0.95; p < 0.05). Again, aside from those from the Kali River and Gaglajur Haor, all of the samples exhibited positive allometric growth patterns (b > 3) with the fish from the Tetulia River being the most positive (b = 3.48). The causes these variations are not yet clearly understood, however; the nutrient availability, environmental variation, and genetic-environmental interactions are likely contributors to the diversities displayed by the hilsa from different regions of Bangladesh.
