Exercise Myokine Irisin Enables Behavioral Stress Resistance in Mice

Physical activity produces widespread benefits to physical and mental health, including protection against stress-related behavioral outcomes. However, the peripheral signals through which physical activity is translated into stress resistance remain incompletely understood. Irisin, an exercise-induced myokine cleaved from the transmembrane protein FNDC5 and released into circulation, has been implicated in improved cognition and neuroprotection, but its role in resistance to behavioral consequences of future adversity has not been examined. Here, we tested whether forced peripheral elevation of irisin is sufficient to protect against stress-induced behavioral outcomes. Adult male C57BL/6 mice received adeno-associated viral (AAV)-mediated peripheral expression of irisin or GFP control. AAV-irisin significantly increased circulating irisin levels, and six weeks later, mice were exposed to inescapable stress, a well-characterized model that produces anxiety-like behaviors, including reducing rodents natural inclination toward sociability. Elevated circulating irisin prevented the stress-induced reduction in social preference without altering general locomotor activity. Moreover, circulating irisin levels positively predicted individual differences in sociability. Additionally, peripheral irisin elevation increased brain-derived neurotrophic factor (Bdnf) expression, which also positively correlated with circulating irisin levels. Finally, we identified expression of the irisin receptor subunit integrin V within the dorsal raphe nucleus, a key brain region implicated in the behavioral outcomes of inescapable stress that is modulated by prior exercise. Together, these findings identify peripheral irisin as a mediator sufficient to confer resistance to future stress, even in the absence of physical activity. Significance StatementPhysical activity protects against the anxiety-like behavioral outcomes of future stress, but the peripheral mediators of this effect remain unclear. Here we show that forced peripheral elevation of circulating irisin, an exercise-induced myokine, is sufficient to produce behavioral stress resistance in mice. Irisin prevented the stress-induced reduction in social preference and positively correlated with individual differences in sociability. Forced expression of peripheral irisin also increased Bdnf expression in the brain, consistent with engagement of central neuroplasticity pathways that support adaptive behavioral responses. These findings demonstrate that an exercise-induced circulating factor can confer resistance to future stress, identifying irisin as a potential prophylactic target for stress-related mental health disorders.

A pilot study on the biological applications of indole alkaloids derived from Nagaland biodiversity

The north-eastern Indian state, Nagaland with its rich biodiversity, is well known for traditional, natural medicines. However, the scientific basis of the working principle(s) of such drugs are yet to be explored. Plant alkaloids, the nitrogen-containing organic molecules, are traditionally been used for various medicinal purposes from ages. There is a need for drug discovery from natural sources to create a broader drug portfolio for human use. And, Nagaland medicinal plant based bioactive molecules have huge potential for the same. As a pilot study, this work aims to study the in silico absorption, distribution, metabolism and excretion (ADME) properties, toxicity prediction and structural properties of the indole alkaloid molecules, Ajmalicine (AJM) and Serpentine (SER) and its docking interaction pattern with various forms of DNA. ADME analysis confirms that the alkaloids meet the criteria of a standard drug. The ProTox II prediction showed the probability of the toxicity profile of the input compounds. Density functional theory (DFT) revealed the HOMO-LUMO energy gap of the drug molecules along with the quantum chemical parameters from the optimized geometry of the ligands. Molecular docking (MD) analysis showed successful binding of the molecules into the groove region of the various forms of DNA structure for both the cases. The results of this theoretical pilot study provides information that will be helpful in the development of effective and modified drugs for medicinal and pharmaceutical purposes as well as in the planning of practical laboratory experiments.

Exome sequencing identifies variants associated with semen quality in Holstein Friesian and Hallikar bulls.

Effective fertility of bulls is dependent on semen quality, often determined based on standard semen evaluation tests. Here we report Whole Exome Sequencing (WES) of 12 bulls from two breeds Holstien Friesian and Hallikar selected based on Ejaculate Rejection Rate (ERR). We explored the possibility of identifying genetic variants from the conserved protein coding regions of genome. A total of 10,510 SNPs and 10.236 INDELs were identified post alignment against reference genome (ARS-UCD 1.2) and were annotated using SnpEff. The number of variants with high and modifier functional impact detected were 145 and 19,122, respectively. Genetic variants common to both high and low ERR group bulls among Holstein Friesian were 08 and in Hallikarthe common variants were 51. Prominent genesviz. UCP2, PANK2, GPD2, PTPRG, LARP7, EZH1, DENND1B and TDRD9 with a role in determining the semen quality were observed to be carriers of the genetic variant.

A miRNAs Based Exploration of promising Biomarkers in Cervical Cancer using Bioinformatic Methods

Cervical Cancer (CC) is a gynecologic cancer. In this cancer early detection is incredibly tough because most of the patients are not have any specific symptoms that results in suspending the proper identification. In this work, we selected TCGA CESC datasets and miRNA Seq analysis was done. The expression profiles of miRNAs in cervical cancer datasets were investigated using bioinformatics tools. The expression profiles of miRNA in Normal tissue, primary tumor and metastatic samples were analyzed. Based on p-value, principal component analysis and comparative literature survey, we reported 6 over-expressed (5X) miRNA at metastatic stage namely, hsa-mir-363, hsa-mir-429, hsa-mir-141, hsa-mir-93, hsa-mir-203b and hsa-mir-18a. Expression profiles were compared in heatmap. The target genes for the selected miRNAs were investigated for interaction and pathway details. The identification of two hub proteins (PTEN and MYC) in Protein-Protein Interaction Network was followed by pathway analysis. Our results indicate that hsa-mir-363, hsa-mir-429, hsa-mir-141, hsa-mir-93, hsa-mir-203b and hsa-mir-18a could be a potential diagnostic biomarkers for early-stage CESC and serve as prognostic predictors for patients with CESC.

Molecular dissection of Carbapenem-resistant Acinetobacter baumannii circulating in Indian hospitals usingWhole Genome Sequencing

ObjectivesCarbapenem-resistant Acinetobacter baumannii (CRAB) has acquired worldwide recognition as a serious nosocomial infection. It poses a concern to hospitalized patients because of the limited therapeutic options available. Thus, we investigated the molecular epidemiology and antibiotic resistance profiles of A. baumannii isolates in India. Materials and MethodsWe characterized 306 retrospective A. baumannii clinical isolates collected from 18 centers across 10 states and 1 Union Territory of India between 2015 and 2019. Molecular epidemiology, and carbapenem resistance were studied by Whole Genome Sequencing. ResultsA total of 105 different Sequence Types (STs) were identified including 48 reported STs and 57 Novel STs. 99 isolates were classified into Clonal Complex 451 (CC451) among which ST848 and ST1956 were the common STs. Carbapenemase resistance was confirmed in all the isolates with the presence of intrinsic blaOXA-51-like genes, and the acquired blaOXA-23 and blaNDM-1 genes. ConclusionMost of the isolates were grouped under clonal complex 451. ST1053 caused an outbreak in Northern India during 2018 and 2019. Novel MLST alleles and STs were also detected, underlining an evolutionary divergence in India. The carbapenem-resistance was dominated by OXA-type carbapenemases and further surveillance of these carbapenem-resistant A. baumannii and antimicrobial stewardship should be strengthened.

A subpopulation of spinocerebellar tract neurons regulates the stability of bipedal stepping

Peripheral and intraspinal feedback is required to shape and update the output of spinal networks that execute motor behavior. We report that lumbar dI2 spinal interneurons of the chick receive synaptic input from afferents and pre-motoneurons. They innervate contralateral premotor networks in the lumbar and brachial spinal cord and their ascending projections innervate the cerebellum. These findings suggest that dI2 neurons function as interneurons in local lumbar circuits and involved in lumbo-brachial coupling and that part of them deliver peripheral and intraspinal feedback to the cerebellum. Silencing of dI2 neurons leads to destabilized stepping in P8 hatchlings with occasional collapses, variable step-profiles and wide-base walking, suggesting that the dI2 neurons may contribute to stabilization of the bipedal gait.

Title - Effects of Gamma Irradiation Against Mating Competitiveness of Male Culex quinquefasciatus (Diptera:Culicidae) in Laboratory

Culex quinquefasciatus is the main vector of lymphatic filariasis in Pekalongan City. Sterile Insect Tehnique (SIT) could be complementary vector control for filariasis. The key success of the technique depend on the ability of laboratory-reared sterile males with the wild-type females. The aim of the research was to determine the mating competitiveness, the fecundity and the fertility of sterile male Culex quinquefasciatus. The pupae of Cx. quinquefasciatus were gamma irradiated at the doses of 60Gy, 70Gy, and 80Gy, while unirradiated pupae were prepared as control. The mosquitoes emerging from the irradiated pupae could mate with a normal female in the cages. It were observed for the mean female laying eggs, the fecundity, the fertility and the mating competitiveness. The data were analyzed by one way ANOVA. The result showed that the irradiated Cx. quinquefasciatus at the doses tested did not affect on the fecundity and the mating competitiveness, but the fertility was disturbed (sterile). A dose of 70 Gy was the optimum dose or a fertility rate of 1.8% (98.2% sterile), and the value of competitiveness (C index) was 0.568. Based on the result, the irradiated Cx. quinquefasciatus can be recommended for semifield application.

A complex containing lysine-acetylated actin inhibits the formin INF2

INF2 is a member of the formin family of actin assembly factors. Dominant mis-sense mutations in INF2 link to two diseases: focal segmental glomerulosclerosis (FSGS), a kidney disease; and Charcot-Marie-Tooth disease (CMTD), a neuropathy. All disease mutations map to the autoinhibitory Diaphanous Inhibitory Domain (DID). Curiously, purified INF2 is not autoinhibited, suggesting the existence of additional cellular inhibitors. We purified an INF2 inhibitor from mouse brain, and identified it as a complex between lysine-acetylated actin (KAc-actin) and cyclase-associated protein (CAP). Inhibition of INF2 by CAP/KAc-actin requires INF2 DID. Treatment of CAP/KAc-actin with histone deacetylase 6 (HDAC6) releases INF2 inhibition, while HDAC6 inhibitors block cellular INF2 activation. INF2 disease mutants are poorly inhibited by CAP/KAc-actin, suggesting that FSGS and CMTD result from reduced CAP/KAc-actin binding. This is the first demonstrated role for lysine-acetylated actin: regulation of an actin assembly factor by a novel mechanism, which we call facilitated auto-inhibition.
